dc.description.abstract | Ctenomys flamarioni e C. minutus são roedores herbívoros de hábito fossorial, que habitam a planície costeira do sul do Brasil, e possuem uma pequena região de simpatria no estado do Rio Grande do Sul. Pouco se sabe a respeito da dieta dos ctenomídeos, sendo comumente sugerido que estes se alimentam de folhas e raízes de gramíneas. Marcadores moleculares de DNA cloroplastidial vêm sendo amplamente aplicados em estudos envolvendo a análise da composição da dieta de herbívoros, principalmente o íntron trnL (UAA), o qual possui uma pequena região conhecida como alça P6, altamente variável e flanqueada por regiões conservadas, permitindo assim a amplificação e identificação de material degradado. O objetivo principal deste trabalho é organizar uma base de dados de sequências do íntron trnL (UAA) de plantas amostradas ao redor das tocas de C. flamarioni e C. minutus e testar o poder de resolução taxonômica da alça P6. Foram selecionados três pontos de coleta; Tramandaí, Torres e Praia do Barco, onde ocorrem respectivamente, C. flamarioni, C. minutus e as duas espécies em simpatria. Para amplificar o íntron trnL (UAA) foram utilizados os primers c e d. Das 91 plantas coletadas foram geradas 73 sequências, totalizando 43 haplótipos, distribuídos em 43 espécies e 41 gêneros. Através do BLAST gerado para cada haplótipo e das árvores filogenéticas constatamos a validade do íntron trnL (UAA) como DNA barcoding de plantas. As análises da alça P6 do íntron trnL (UAA) demonstraram que esta região apresenta grande potencial na identificação de material degradado e análise da dieta de herbívoros. | pt_BR |
dc.description.abstract | Ctenomys flamarioni and C. minutus are herbivorous rodents with fossorial habit, which inhabit the southern Brazilian coastal plain, and have a small area of sympatry in the state of Rio Grande do Sul. Little is known about the diet composition of ctenomyids and it is commonly suggested that they feed on leaves and roots of grasses. Through the development of molecular markers of chloroplast DNA, such as trnL (UAA) intron, it has been possible to assess more accurately the diet of herbivores. This intron has a small region known as P6 loop that has been used in the identification of degraded material and proved to be effective in the analysis of the diet of herbivores. The main objective of this work is to organize a database of sequences of trnL (UAA) intron of the plants found around the burrows of C. flamarioni and C. minutus and to test the taxonomic resolution power of the P6 loop. We selected three sampling sites, Tramandaí, Torres and Praia do Barco where occurs respectively C. flamarioni, C. minutus and these two species in sympatry. To amplify the trnL (UAA) intron we used the primers c and d. Of the 91 plants collected, were generated 73 sequences, totalizing 43 haplotypes distributed among 43 species and 41 different genera of plants. Through the BLAST carried by each haplotype and by the phylogenetic trees reconstructions we were able to verify the validity of the trnL (UAA) intron as DNA barcoding of plants. The analyses performed with the P6 loop demonstrated a good taxonomic resolution demonstrating that this region has great potential to identify degraded material and also to analyze the diet of herbivores. | en |
